Application of hight-performance liquid chromatography in assisted reproduction

Abstract:

Cílem této práce bylo (a) zavést metody pro stanovení vitaminů a antioxidantů v seminální plazmě mužů a porovnat jejich hladiny mezi různými skupinami pacientů a (b) zavést metody pro stanovení aminokyselin a oxokyselin v kultivačním médiu pro porovnání metabolismu mezi jednotlivými lidskými embryi a nalézt vztah mezi metabolismem aminokyselin a morfokinetickými parametry embrya určenými time-lapse analýzou. Byly zavedeny dvě metody pro simultánní stanovení antioxidantů (vitaminů) v seminální plazmě mužů, jedna pro stanovení kyseliny askorbové a močové, druhá pro stanovení retinolu a ?-tokoferolu. V obou případech se jedná o metody vysokoúčinné kapalinové chromatografie s UV detekcí. Vzorky spermatu byly získány od mužů léčících se pro neplodnost. Při stanovení kyseliny askorbové a močové, byly ejakuláty odstředěny a seminální plazma naředěna roztokem dithiothreitolu, po filtraci byly vzorky dávkovány na analytickou kolonu s vázanou reverzní fází. Při stanovení retinolu a ?-tokoferolu byla seminální plazma deproteinována ethanolem, vitaminy byly extrahovány n-hexanem a extrakty odpařeny do sucha v atmosféře dusíku. Odparek byl rozpuštěn v ethanolu a po filtraci byl vzorek dávkován na analytickou kolonu s vázanou reverzní fází. Analytické parametry těchto metod byly vyhovující. Nebyly nalezeny žádné statisticky významné rozdíly v koncentracích kyseliny askorbové i močové mezi kuřáky a nekuřáky [327,4 (263,0) ?mol/l a 298,7 (146,8) ?mol/l vs. 400,0 (367,8) ?mol/l a 304,2 (138,2) ?mol/l, p > 0,05]. Podobně hladiny retinolu a ? tokoferolu se statisticky významně nelišily mezi kuřáky a nekuřáky [12 (6) nmol/l a 1,9 (0,3) ?mol/l vs. 13 (8) nmol/l a 1,8 (0,5) ?mol/l, p > 0,05]. Byly zavedeny dvě metody vysokoúčinné kapalinové chromatografie s fluorescenční detekcí, jedna pro stanovení vybraných aminokyselin, druhá pro stanovení vybraných oxokyselin v kultivačním médiu. Dvacet čtyři hodin po intraplazmatické injekci spermií, byla jedna, náhodně vybraná zygota, přemístěna do kultivačního média. Po kultivaci byla kultivační média, ve kterých se inkubovala embrya, a média, ve kterých se embrya neinkubovala (slepý vzorek), uchována při 80 °C. Vzorky médií byly derivatizovány (a) ortho-ftalaldehydem (stanovení aminokyselin) a (b) ortho fenylendiaminem (stanovení oxokyselin) za vzniku vysoce fluoreskujících derivátů. Nebyly pozorovány žádné statisticky významné rozdíly v produkci/spotřebě a obratu aminokyselin mezi dělícími se a nedělícími se embryi. Hladiny aminokyselin, které jsou součástí média, nekorelovaly s morfokinetickými znaky embrya, s výjimkou stupně fragmentace embrya ve 3. dni po oplození, kde byla pozorována negativní korelace s hladinou kyseliny glutamové a pozitivní s hladinami glutaminu, glycinu a taurinu. V médiu některých dělících se i nedělících se embryí se objevily esenciální aminokyseliny, jejichž hladiny spolu navzájem korelovaly, ale nekorelovaly s hladinami aminokyselin, které jsou součástí kultivačního média. V médiích před inkubací i po inkubaci lidských embryí byly pozorovány pouze hladiny pyruvátu a 2-oxoglutarátu.

Abstract:

The aim of this work was (a) to develop methods for the determination of vitamins and antioxidants in human seminal plasma and to compare their levels between different groups of patients and (b) to develop methods for the determination of amino acids and ?-keto acids in human culture medium and to compare amino acids metabolism between human embryos and to find some relationship between amino acids metabolism and the embryo morphokinetic parameters studied via time-lapse analysis. Two methods for the simultaneous determination of antioxidants (vitamins) in human seminal plasma were developed; one for the measurement of ascorbic acid and uric acid and the second for the determination of retinol and ?-tocopherol, both using an high-performance liquid chromatography with UV detection. Semen samples were obtained from consecutive male partners of couples presenting for a fertility evaluation. For the determination of ascorbic acid and uric acid, ejaculates were centrifuged and the seminal plasma diluted with dithiothreitol solution and after a filtration injected onto an analytical reverse-phase column. For the determination of retinol and ?-tocopherol, seminal plasma was deproteinized with ethanol, vitamins were extracted with n-hexane and extracts were evaporated to dryness, under nitrogen. The dried residue was resuspended in ethanol and after a filtration, injected onto an analytical reverse-phase column. Analytical performance of these methods was satisfactory. No statistically significant differences in both ascorbic acid and uric acid concentration between the smokers and non-smokers [327.4 (263.0) ?mol/l a 298.7 (146.8) ?mol/l vs. 400.0 (367.8) ?mol/l a 304.2 (138.2) ?mol/l, p > 0,05] were observed and similarly no statistically significant differences in both retinol and ?-tocopherol concentration between the smokers and nonsmokers [12 (6) nmol/l a 1.9 (0.3) ?mol/l vs. 13 (8) nmol/l a 1.8 (0.5) ?mol/l, p > 0,05] were found. Two methods, one for the determination of selected amino acids and the second for the determination of keto acids in culture medium using high-performance liquid chromatography with fluorescence detection, were developed. Twenty hours after the intra-cytoplasmic sperm injection, one randomly selected zygote was transferred to the culture medium. After incubation, culture medium, in which the embryo was incubated, and blank medium were immediately stored at 80 °C. Medium samples were derivatized (a) with ortho-phthalaldehyde (determination of amino acids) and (b) with ortho-phenylenediamine (determination of keto acids) to forms highly fluorescent amino (?-keto) acid derivatives. For the separation, a reverse-phase column was used and gradient elution was carried out. Analytical performance of these methods was satisfactory. No statistically significant differences between dividing and arresting embryos were observed in terms of amino acids production/consumption and turnover. Amino acids which were part of the culture medium did not exhibit any statistically significant correlation with morphokinetic parameters, with the exception of the grade fragmentation on day 3; there were negative correlation with glutamate, and positive with glutamine, glycine and taurine. In some dividing and in some arresting embryos appeared new (essential) amino acids which strongly correlated with each other, with methionine, but not with any other amino acid that is a regular part of the culture medium. Only pyruvate and ?-ketoglutarate were observed in culture medium before and after incubation of human embryos.
