Vztah lipidů ušního mazu a krevního séra

Relation of cerumen and serum lipids

Abstract:

Předložená diplomová práce se zabývá problematikou ušního mazu ve vztahu ke krevnímu séru. Je diskutována závislost pěti lipidových frakcí cerumina (fosfolipidy, diacylglyceroly, volné mastné kyseliny, triacylglyceroly a estery cholesterolu) na lipidech krevního séra a dalších sledovaných znacích (fosfolipidy, diacylglyceroly, volné mastné kyseliny, triacylglyceroly a estery cholesterolu v krevním séru, věk, váha, výška, body mass index a dekáda). Separace lipidových frakcí z jednotlivých vzorků získaných od 33 jedinců byla prováděna metodou tenkovrstevné chromatografie. Pomocí statistické analýzy vícerozměrných dat byly určeny shluky podobných znaků a subjektů s využitím metody hlavních komponent, faktorové analýzy a analýzy shluků. Byly nalezeny vzájemné korelace mezi znaky popisujícími lipidy krevního séra a mezi znaky charakterizujícími tělesnou konstituci jedince. K určení vzájemných skrytých vazeb jak mezi znaky, tak i mezi subjekty byla zařazena kanonická korelační analýza, která odhalila lineární závislost mezi pěticí lipidových složek ušního mazu a desítkou ostatních sledovaných znaků. Vzhledem ke zjištěné lineární závislosti byl prostřednictvím vícerozměrné lineární regrese určen vliv jednotlivých sledovaných znaků na lipidy ušního mazu. Bylo odhaleno, že hladina diacylglycerolů v ceruminu je přímo úměrná hladině fosfolipidů krevního séra a snižuje se s věkem. Množství volných mastných kyselin v mazu je přímo úměrné hladině volných mastných kyselin a esterů cholesterolu v krevním séru, body mass indexu a výšce, jejich množství má klesající tendenci ve vztahu k tělesné hmotnosti. Obsah esterů cholesterolu v mazu je přímo úměrný hladině volných mastných kyselin a esterů cholesterolu v krevním séru a nepřímo úměrný množství krevních triacylglycerolů. Hodnoty esterů cholesterolu se zvyšují s věkem. Nebylo zjištěno ovlivnění fosfolipidů a triacylglycerolů v ušním mazu žádnými ze sledovaných znaků. Práce odhalila nové informace o faktorech ovlivňujících složení ušního mazu a přispěla tak k rozšíření znalostí o fyziologii ušního mazu.

Abstract:

The thesis concerns with dependence of five earwax lipid fractions (phospholipids, diglycerides, free fatty acids, triglycerides and cholesterol esters) on serum lipids and other monitored compounds and items forming here the variables (phospholipids, diglycerides, free fatty acids, triglycerides and cholesterol esters in serum, age, weight, height, body-mass index and decade). Separation of lipids fraction was performed by the thin layer chromatography. In the thesis there were analysed 33 subjects. Using the multidimensional statistic analysis, the cluster of similar variables and cluster of similar subjects were treated. The exploratory analysis was performed by the principal components analysis, an also with the factor analysis and the cluster analysis. The correlations between monitored factors were found. The canonic correlation analysis revealed linear dependence among groups of five dependent variables (earwax lipid levels) and ten independent variables (the rest of monitored factors). Because to founded linear dependence, the effect of individual watched variables on ear wax´s lipids has been analysed with the multidimensional linear regression. It was revealed that the level of diglycerides in cerumen is directly proportional to the level of serum phospholipids but it gets down with age. The number of free fatty acids in the ear wax is directly proportional to the level of free fatty acids and cholesterol esters in serum, body mass index and height. Their level has a falling tendency in the relationship on the physical weight. The contents of cholesterol esters in ear wax is directly proportional to the level of free fatty acids, cholesterol esters in serum and inversely proportional to the number of blood triglycerides. The cholesterol esters values are increasing with age. No influence of any monitored factors was observed on the level of an actual phospholipids and triglycerides in ear wax. The thesis reveals the new information about factors influencing a composition of the ear wax and helps into the extension our knowledge about ear wax physiology.
