Modern approaches to classification, diagnosis and therapy of hematological malignancies

Abstract:

Bakalářská práce se zabývá klasifikací a diagnostikou hematologických malignit s využitím nejmodernějších technik. Práce je rozdělena do pěti hlavních kapitol. Nejprve stručně popisuje vznik hematologických malignit. Následuje kapitola zabývající se nejmodernějším klasifikačním systémem hematologických malignit - WHO klasifikace, která je neustále doplňována o nové informace. WHO klasifikace k tomuto využívá nejnovější laboratorní metody, a to především z oblasti molekulární genetiky. Třetí kapitola je věnována incidenci a prevalenci hematologických malignit. Obsahuje informace o nejčastějších typech hematologických malignit, počtu nově diagnostikovaných případů a úmrtnosti na jednotlivých kontinentech v důsledku těchto nádorových onemocnění. Dále se práce zabývá moderními diagnostickými metodami, využívanými v klinické praxi. Zároveň je zde uveden i relativně nový obor: proteomika a s ní spojené instrumentální techniky, které prozatím mají uplatnění v rámci experimentální praxe. V kapitole o diagnostice jsou zahrnuty i molekulární biomarkery a jejich využití pro diagnostiku těchto onemocnění. Poslední kapitola pojednává o současných trendech terapie hematologických malignit s využitím inhibitorů buněčné signalizace, specifických protilátek a transplantace krvetvorných kmenových buněk.

Abstract:

Bachelor thesis is focused on modern approaches to classification, diagnosis and therapy of haematological malignancies. There are five major chapters: pathogenesis, classification, incidence and prevalence, diagnostic techniques and therapy of haematological malignancies. The first part briefly describes the biological mechanism of haematological malignancies. The WHO classification, the modern classification system of haematological malignancies, is described in the second part. The information about the most common types of haematological malignancies, their incidence, prevalence, numbers of diagnosed patients and mortality is in the chapter three. The fourth chapter deal with diagnostic techniques used in clinical practise, such as flow cytometry, polymerase chain reaction or DNA sequencing. The proteomics, the new field of research in malignancies is also described here. The final part concerns on treatment. New inhibitors, monoclonal antibodies and allogeneic haematopoietic stem cell translation are described.
