Molecular mechanisms of resistence to thiosemicarbazones

Abstract:

Thiosemicarbazony jsou díky schopnosti snižovat hladinu iontů železa slibnými terapeutiky v nádorové terapii. Potenciálně jsou využitelné také v léčbě neuroblastomů, nejčastějších solidních tumorů dětského věku. Inhibiční vliv thiosemicarbaznů na nádorové buňky byl opakovaně pozorován, přesto zůstává specifická subpopulace, která je vůči jejich působení do značné míry rezistentní. Potenciálním vysvětlením je výskyt nádorových kmenových buněk se zvýšenou tolerancí k thiosemicarbazonům, které se následně podílí na vzniku relapsů a metastáz. Cílem práce je tvorba a charakterizace této rezistentní populace buněk a její porovnání s neovlivněnou buněčnou populací. Práce se soustředila na detekci exprese vybraných markerů, souvisejících s nádorovými kmenovými buňkami, nebo úzce spojených s mechanismem působení thiosemicarbazonů. Dále byly využity některé funkční testy pro posouzení kmenovosti a tumorigeneze. Výsledky práce ukazují, že změny v expresi markerů kmenovosti navozené u rezistentní subpopulace neodpovídají zvýšení charakteru kmenovosti. Přesto funkční testy naznačují udržení kmenového charakteru, na jehož zachování může mít významný podíl HIF-2α.

Abstract:

Thanks to their ability to lower the level of iron ions, thiosemicarbazones are a promising therapeutic in tumor treatment. They could potentially also be used in the treatment of neuroblastoma, most commonly in solid tumors in children. The inhibitive influence of thiosemicarbazones onto cancer cells has been repeatedly observed, however there still remains a subpopulation of those which are substantially resistant. This could potentially be explained by the existence of cancer stem cells with higher resistance to thiosemicarbazones, which contribute to the creation of relapses and metastasis. This research aims to create a characterisation of this resistant population of cells, as well as to compare them with a population of unaffected control cells. The research conducted is focused on the detection of expression of chosen markers linked to the tumorous stem cells, or closely related to the mechanism of how the thiosemicarbazones work. Furthermore, certain functional tests were used to determine familiarity and tumorigenesis. The results of the research show that the changes in the expression of stem cell group markers in the resistant population belie the heightened character of the stemness. However, the practical tests still suggest a continuation of stemness, which might be in large part due to HIF 2α.
