Evaluation of flavonoid mutagenicity by Ames test

Abstract:

Diplomová práce se v teoretické části věnuje současným přístupům a trendům v hodnocení genotoxicity s detailním zaměřením na Amesův test. Charakterizuje flavonoidní látky s ohledem na jejich genotoxickou a biologickou aktivitu. Experimentální část práce se zabývá hodnocením mutagenity tří flavonoidů - naringeninu, genisteinu a quercetinu Amesovým testem bez a s metabolickou aktivací in vitro. Standardní látkou sloužící jako pozitivní kontrola byl nitrofurantoin. K detekci mutagenity bylo využito indikátorových bakteriálních kmenů Salmonella typhimurium His- TA98 a TA100. Výsledky všech testů byly hodnoceny statistickým programem SALM. Jediným prokázaným mutagenem v Amesově testu byl flavonoid quercetin. Flavonoidy naringenin a genistein vykazovaly při vyšších koncentracích toxický efekt. Výsledky ukazují, že i přes řadu zdraví prospěšných aktivit flavonoidů je důležité hodnotit jejich potenciální rizika pro lidský organismus, zejména při jejich zvýšeném obsahu ve stravě a potravinových doplňcích.

Abstract:

The diploma thesis, in theoretical part, deals with current approaches and trends in the assessment of genotoxicity with a detailed focus on the Ames test. It characterizes flavonoid substances with regard to their genotoxic and biological activity. The experimental part of the work deals with the evaluation of the mutagenicity of three flavonoids - naringenin, genistein and quercetin using the Ames test with and without metabolic activation in vitro. The standard substance serving as a positive control was nitrofurantoin. The indicator bacterial strains Salmonella typhimurium His- TA98 and TA100 were used to detect mutagenicity. The results of all tests were evaluated by the statistical program SALM. The only proven mutagen in the Ames test was the flavonoid quercetin. The flavonoids naringenin and genistein showed a toxic effect at higher concentrations. The results show that despite the many health-promoting activities of flavonoids, it is important to evaluate their potential risks for the human body, especially when their content is increased in the diet and food supplements.
