Analysis of microRNAs involved in regulation of glioblastoma stem cells

Abstract:

Multiformní glioblastom (GBM) je nejčastější primární nádor mozku astrocytárního původu. Prognóza pacientů s tímto onemocněním je i přes maximálně možnou v současné době dostupnou konvenční terapii velmi nepříznivá. Důvodem je především časně se vyskytující relaps onemocnění, jehož často diskutovanou příčinou je přítomnost glioblastomových kmenových buněk (GSCs). Jedná se chemoradiorezistentní subpopulaci nádorových buněk s tumoriniciační kapacitou, jež se v mnoha ohledech podobá zdravým kmenovým buňkám (SCs). Byla u ní tedy zaznamenána exprese stejných markerů kmenovosti a aktivita specifických signálních drah jako je tomu právě u SCs. Jedněmi z klíčových molekul ovlivňujícími celou řadu buněčných procesů včetně udržování kmenového fenotypu jsou mikroRNA (miRNA). Tyto krátké endogenně se exprimující nekódující RNA posttranskripčně regulují genovou expresi a jsou tedy významně zapojeny do regulace mnoha signálních drah nevyjímaje signalizace spojené právě s biologií GSCs. Detailnější studium jejich funkce v GSCs proto může vést k objevení nových diagnostických markerů a terapeutických cílů a zlepšit tak celkovou prognózu pacientů s GBM. Cílem této práce bylo odvodit párové primární buněčné linie GSCs a non-GSCs a analyzovat u nich expresi kmenových markerů. Následně byla u těchto buněk provedena globální expresní analýza miRNA profilů, přičemž vybrané, mezi oběma skupinami významně deregulované miRNA, pak byly studovány in vitro pomocí sphere formation assay. Nakonec byla rovněž prozkoumána exprese miRNA deregulovaných u GSCs ve vztahu k jejich expresi a údaji o přežívání u pacientů s GBM za účelem odhalení miRNA s prediktivním a prognostickým potenciálem.

Abstract:

Glioblastoma multiforme (GBM) is the most frequent primary brain tumor of astrocytic origin. The prognosis of patients with this disease is very unfavourable despite the maximum possible and currently available conventional therapy. The primary reason for this is the early occurrence of relapse which is often thought to be caused by the presence of glioblastoma stem cells (GSCs). They comprise chemoradioresistant subpopulation of cancer cells with tumor-initiating capacity which is in many ways similar to healthy stem cells (SCs). Just like SCs, it is characterized by the expression of the same markers of stemness and the acitivity of specific signalling pathways. One group of key molecules affecting a whole range of cellular processes, including the maintenance of stem phenotype, are microRNAs (miRNAs). These short endogenously expressed non-coding RNA post-transcriptionally regulate gene expression and thus are significantly involved in the regulation of many signalling pathways including those related to GSC biology. More detailed study of their function in GSCs may, therefore, lead to the discovery of new diagnostic markers and therapeutic targets and improve the overall prognosis of patients with GBM. The aim of this thesis was to derive paired primary cell lines (GSCs and non-GSCs) and analyze their expression of stemness markers. A subsequent global expression analysis of miRNA profiles was performed on these cells and selected miRNAs significantly dysregulated between both groups were then studied in vitro by sphere formation assay. Finally, the expression of miRNAs dysregulated in GSCs was reviewed in their relation to survival parameters of patients with GBM in order to discover miRNAs with predictive and prognostic potential.
