Expresní profily buněčných linií embryonálních nádorů při aplikaci kombinované diferenciační terapie

Expression profiles of the embryonal tumor cell lines after application of the combined differentiation therapy

Abstract:

Retinoidy jsou známy jako efektivní induktory buněčné diferenciace, a to jak v podmínkách in vivo, tak in vitro. Díky této schopnosti jsou retinoidy často používány v protinádorové terapii. Předkládaná disertační práce je zaměřena na studium možností zesílení antineoplastického efektu kyseliny all-trans retinové pomocí kombinované aplikace s inhibitory lipooxygenáz (LOX) a cyklooxygenáz (COX). V této studii byly používány kyselina kávová jako inhibitor 5-lipoxygenázy a celecoxib jako inhibitor cyklooxygenázy-2. Jako biologické modely byly vybrány dvě neuroblastomové (SK N BE(2) a SH-SY5Y) a dvě smeduloblastomové (Daoy a D283 Med) sbírkové buněčné linie; část experimentů byla prováděna rovněž na 4 pacientských meduloblastomových liniích. Pro monitorování změn v expresi 440 genů funkčně spojených s biologií nádorů byly použity nízkohustotní oligonukleotidové mikročipy Human Cancer Oligo GEArray (SABiosciences). Změny exprese vybraných kandidátních genů (CDKN1A, GDF15, HLA-C, HMGA1, TIMP1) byly následně potvrzeny pomocí RT PCR. Analýza detekovaných změn genové exprese prokázala zvýšení exprese genů zapojených v procesech retinoidy indukované diferenciace v závislosti na koncentraci použitých činidel; zejména se jednalo o geny obecně spojované s buněčnou diferenciací, geny podílející se na regulaci přestaveb cytoskeletu, geny zapojené do metabolismu mitochondrií atd. Celkově byla prokázána vyšší senzitivita neuroblastomových linií ke kombinované aplikaci ATRA s kyselinou kávovou, resp. s celecoxibem. Získané výsledky podporují naši původní hypotézu, že buněčná diferenciace indukovaná ATRA může být zesilována kombinací s LOX, resp. COX inhibitory.

Abstract:

Retinoids are known to be effective inducers of cell differentiation under both in vivo and in vitro conditions. Due to this capability, retinoids are often used in recent antitumor therapy. This Ph.D. thesis is aimed to investigate possibilities of enhancement of the ATRA-induced antineoplastic effects by combined treatment with inhibitors of lipoxygenases (LOX) or cyclooxygenases (COX). Caffeic acid as an inhibitor of 5 lipoxygenase and celecoxib as an inhibitor of cyclooxygenase-2 were chosen for this study. Two neuroblastoma (SK-N-BE(2) and SH-SY5Y), and two medulloblastoma (Daoy and D283 Med) established cell lines were chosen as biological models; some experiments were also performed using 4 in-house medulloblastoma cell lines. The Human Cancer Oligo GEArray low-density oligonucleotide microchips (SABiosciences) were used to evaluate changes in expression of 440 cancer-related genes. Changes in expression of selected candidate genes (CDKN1A, GDF15, HLA-C, HMGA1, TIMP1) were then confirmed by RT-PCR.Analysis of detected changes in gene expression showed a concentration-dependent increase in expression of genes known to be involved in the process of retinoid-induced differentiation; especially of genes generally associated with cell differentiation, genes participating in regulation of cytoskeleton rearrangements, genes involved in mitochondrial metabolism, etc. To summarize, results also indicated higher sensitivity of neuroblastoma cell lines to the combined treatment with ATRA and caffeic acid or celecoxib. Obtained results support our initial hypothesis that ATRA-induced cell differentiation could be enhanced by its combination with LOX/COX inhibitors.
