Identification of Treponema pallidum sigma factor-dependent promoters

Abstract:

Sigma faktor je disociabilní podjednotkou bakteriální RNA polymerázy a je stěžejní pro iniciaci transkripce. V bakteriích je přítomno několik sigma faktorů a každý má svou cílovou skupinu genů, u kterých řídí transkripci. Treponema pallidum subsp. pallidum, původce syfilis, má pět sigma faktorů, cílové geny však nejsou známy. Jelikož nelze kultivovat T. pallidum in vitro, pro studium treponemálních genů byla využita Escherichia coli, která nese čtyři sigma faktory homologické k treponemálním. Práce sledovala expresi genů obsažených v genomové knihovně T. pallidum a expresi z promotorů získaných klonováním treponemální DNA. Studium exprese probíhalo v kmenech E. coli deficientních pro sigma faktory a při nadprodukci treponemálních sigma faktorů za využití qRT-PCR a luminiscence. Byla potvrzena transkripce genu polA sigma faktorem RpoE, která byla již dříve nalezena u T. pallidum. Byla popsána úloha sigma faktoru RpoN v transportu, kde jako cílové geny byly identifikovány yaeE a abc, kódující transportér aminokyselin, a ptsN-1, kódující fosfotransferázový systém.

Abstract:

Sigma factor is a dissociable subunit of bacterial RNA polymerase and is vital for transcription iniciation. There are several sigma factors present in bacteria and each one has its own set of target genes where it controls transcription. Treponema pallidum subsp. pallidum, the causative agent of syphilis, possesses five sigma factors. However, target genes remain unknown. As T. pallidum is nonculturable in vitro, Escherichia coli was exploited to study T. pallidum genes. E. coli bears four sigma factors which are homologous to the sigma factors of T. pallidum. The thesis examined gene expression of T. pallidum genes from a genomic library and expression from promoters gained by cleavage of treponemal DNA. Expression studies were conducted in E. coli sigma factor-deficient strains and during sigma factor overexpression using qRT-PCR and luminescence measurements. Transcription of the polA gene by RpoE was confirmed as stated formely for T. pallidum. RpoN function in transport was identified, the target genes included yaeE and abc, encoding amino acid transporter, and ptsN-1 coding for phosphotransferase system.
