The determination of selected serum amino acids in patients with liver disease

Abstract:

Práce je věnována stanovení aminokyselin u pacientů s jaterním onemocněním. Játra se podílejí, kromě jiného i na metabolismu aminokyselin. Byla vyvinuta HPLC metoda pro stanovení aminokyselin v séru. Před separací byly aminokyseliny derivatizovány naftalenem-2,3-dikarboxaldehydem. Pro separaci byla použita chromatografická kolona Discovery? Bio Wide Pore C18, 15 cm x 4,6 mm o velikosti částic 5 m. Deriváty byly monitorovány fluorescenčním detektorem. Mobilní fází A byla směs 10mmol/l NaH2PO4 a ethanolu v poměru 80:20, pH 7,2 ? 0,1 a mobilní fází B byla směs 10mmol/l NaH2PO4 a ethanolu v poměru 50:50, pH 7,2 ? 0,1. Analytické parametry metody u vybraných aminokyselin byly vyhovující. Linearita byla dosažena v celém rozsahu testovaných koncentrací. U pacientů s jaterním onemocněním byly v porovnání s kontrolní skupinou nalezeny statisticky významně vyšší hladiny histidinu, citrullinu, tyrosinu, methioninu a tryptofanu a nižší hladiny argininu.

Abstract:

This work deals with the determination of amino acids in patients with liver disease. Liver is involved, among other things the metabolism of amino acids. HPLC method for the determination of amino acids in serum was developed. Before the separation, amino acids were derivatized with naphthalene-2,3-dicarboxaldehyde. For separation was used chromatographic column Discovery? Bio Wide Pore C18 column, 15 cm x 4.6 mm with a particle size of 5 m. The derivatives were monitored by a fluorescence detector. Mobile phase A was a mixture of 10mmol/L NaH2PO4 and ethanol (80:20, v/v), pH 7.2 ? 0.1 and mobile phase B was a mixture of 10mmol/L NaH2PO4 and ethanol (50:50, v/v) pH 7.2 ? 0.1. Analytical parameters of method for the determination of selected amino acids were satisfactory. Linearity was achieved in the whole range of tested concentrations. Amino acid levels of patients with liver disease and a control group were compared with found significantly higher levels of histidine, citrulline, tyrosine, methionine and tryptophane and lower levels of arginine.
