Hydra as potential model organism in stem cells and cancer stem cells research

Abstract:

Kmenové buňky a jejich biologie představují jednu z nejdynamičtěji se rozvíjejících oblastí vědeckého výzkumu zasahujícího do mnoha různých odvětví. Před několika desítkami let byl představen koncept nádorových kmenových buněk, jehož hlavní myšlenka spočívá v tom, že malá skupina buněk je zodpovědná za růst a přežívání nádoru a často také jeho rezistenci k terapii či případný relaps. Identifikace a eradikace těchto buněk a analýza jejich niky ovšem není tak jednoduchá, jak se původně doufalo. Z těchto důvodů jsou vyhledávány jednoduché modelové organismy, které by detailnějšímu pochopení molekulárních pochodů kmenových a nádorových kmenových buněk mohly posloužit. Díky své stavbě a vlastnostem se pro tento účel jeví být vhodní malí sladkovodní živočichové ze skupiny nezmarů. Tato bakalářská práce se zaměřuje na shrnutí poznatků týkajících se těchto organismů a jejich kmenových buněk s ohledem na molekulární signalizaci a souvislosti s lidskými nádorovými kmenovými buňkami a potenciálním využitím jako modelových organismů pro studium kmenových a nádorových kmenových buněk. V práci je obsažena také kapitola o doposud zaznamenaném výskytu nádorů u nezmarů.

Abstract:

Stem cells and their biology are one of the most dynamically developing areas of scientific research and influence many different fields. A few decades ago, the concept of cancer stem cells was introduced, the main idea of which is that a small group of cells is responsible for the growth and survival of the tumor and often also its resistance to therapy or potential relapse. Identifying and eradicating these cells and analyzing their niche is, however, not as simple as hoped. For these reasons, simple model organisms are sought to serve as a vessel for a more detailed explanation of the stem and cancer stem cells molecular processes. Small freshwater organisms from the genus Hydra are increasingly being considered as model organisms for the study of these cells due to their body structure and properties. This bachelor thesis focuses on the summarization of knowledge using these organisms and their stem cells with respect to molecular signaling and connections with human cancer stem cells. It, additionally, concentrates on the organisms’ effective use as model organisms for the study of stem and cancer stem cells. The work also contains a chapter on tumors in Hydra observed so far.
