Bacteriocins of grampositive bacteria and their detection

Abstract:

Tato diplomová práce se zabývá bakteriemi mléčného kvašení a jejich produkcí bakteriocinů. Jsou zde popsány jednotlivé mléčné bakterie a fermentované mléčné výrobky, ve kterých se tyto bakterie používají. Dále se tato práce podrobněji věnuje bakteriocinům bakterií mléčného kvašení, jejich spektru účinnosti, mechanizmu působení a možnostem jejich využití. Nakonec je zmíněn princip polymerázové řetězové reakce (PCR), což je jedna z nejvyužívanějších metod molekulární biologie. Technika PCR se používá k detekci a identifikaci bakterií mléčného kvašení i bakteriocinů. V praktické části byla provedena izolace a identifikace bakterií mléčného kvašení a stanovena produkce bakteriocinů, a to jak klasickými metodami, tak i pomocí metody PCR.

Abstract:

This thesis deals with lactic acid bacteria and thein bacteriocin production. Individual lactic acid bacteria and fermented dairy products are described in this work. Further, this study targets the bacteriocins of lactic acid bacteria, their spectrum of activity, mechanism of action and possibilities of their application. Finally, principle of polymerase chain reaction (PCR), one of the most used method of molecular biology, is mentioned. The technique PCR is commonly applied for detection and identification of lactic acid bacteria and also bacteriocins. In practical part there was performed isolation and identification of lactic acid bacteria and determined formation of bacteriocins, namely both by classical methods and by PCR method.
