Bioinformatics methods to connect the methylation data from profiling microarrays and next-generation sequencing

Abstract:

This diploma thesis investigates the relationship between methylation levels mea-sured by Illumina Infinium 450K or EPIC microarrays and NGS sequencing of bisulfi-te-converted molecules, two popular experimental techniques. The aim is to un-derstand how to convert the methylation data measured by these two different me-thodologies. A third-order polynomial regression model emerged as the most accu-rate in predicting DNA methylation beta-values, as evidenced by its high R-squared values and lower MOR values. Despite some discrepancies in sample classification, a high level of similarity was observed between the scores assigned to microarray data and converted beta-values, demonstrating the effectiveness of the conversion model. The study concludes with the development of a model for converting methy-lation data from NGS bisulfite sequencing to approximate microarray values, which could potentially be utilized for tumor classification and further epigenetic studies.
