Lowering expression of apurinic/apyrimidinic endonuclease APE1 in human embryonic stem cells and its effect on their self renewal and differentiation potential

Abstract:

Lidské embryonální kmenové buňky by se měly, díky své schopnosti proměnit se v kterýkoliv buněčný typ, v blízké budoucnosti stát základem léčby onemocnění a poranění, kterým součaná medicína neumí čelit. Jednou z hlavních překážek, které zatím brání využití těchto buněk v tzv. buněčné terapii, je nestabilita jejich genomu. Během dlouhodobého udžování embryonálních buněk in vitro dochází k přestavbám DNA, jejichž nahromadění jsou v buňkách in vivo asociována s rozvojem nádorových onemocnění. Vznik genomových abnormalit je přitom spojován se samotnou dlouhodobou kultivací in vitro a nedokonale nastavenými podmínkami tohoto procesu. Bylo prokázáno, že v průběhu dlouhodobé kultivace in vitro signifikantně poklesá především aktivita bázově excizní opravy, jednoho z hlavních DNA opravných mechanismů, které udržují genom a chrání ho před poškozením, a to v závislosti na poklesu hladiny proteinu APE1, zahrnutého do mechanismu této opravy. Absence reparace, kterou bázově excizní oprava zajišťuje, by proto mohla úzce souviset se vznikem poškození genomu. Ve své diplomové práci jsem ověřil podstatu snižení úrovně bázově excizní opravy, dále jsem zkoumal možný vliv tohoto reparačního mechanismu na poškození genomu a pokusil jsem se také chemicky modulovat úroveň APE1 v lidských embryonálních kmenových buňkách.

Abstract:

Human embryonic stem cells should become in the near future a basis of treatment of injuries and illnesses where the current medicine is helpless due to their ability to differentiate themselves into any cell types. One of the main obstacles for their current use in the field of cell therapy is their genome instability. During a long term in vitro cultivation of embryonic cells, restructuring of DNA occurs, cumulating of which is associated with tumour origin in cells in vivo. Genome abnormalities origin is concurrently connected to the long-term in vitro cultivation and imperfectly set conditions of this process. It has been demonstrated that in the course of a long term in vitro cultivation particularly the activity of base excision repair, one of the main DNA corrective mechanisms maintaining genome and protecting it from damage, significantly decreases in dependency on decrease of the level of protein APE1, which is included in this corrective way. Absence of reparation, which base excision repair assures, could be then closely connected to genome damage origin. In this diploma thesis the basis of the decrease of the base excision repair level has been verified, then a possible influence of this reparation mechanism to genome damage has been studied and also a chemical modulation of the level of APE1 in human embryonic stem cells has been performed.
