Solving protein structure using X-ray diffraction

Abstract:

Tato bakalářská práce se věnuje rentgeno-strukturní analýze, které se zde využívá ke zjištění struktury zkoumané ornithin aminotransferázy (OAT), pocházející z hrachu setého. Byla exprimována, jako rekombinantní protein, v Escherichia Coli. Druhým zkoumaným proteinem je lektin CV-IIL z organismu Chromobacterium violaceum, exprimovaného taktéž v Escherichia Coli. U ornithin aminotransferázy (OAT) je práce zaměřena na optimalizaci purifikace a následný výběr vhodného prostředí pro krystalizaci tohoto enzymu a sběr vysoko rozlišených difrakčních dat u lektinu CV-IIL, jde o řešení struktury jednobodového mutanta (v pozici 22 nahrazen serin alaninem)za pomoci molekulárního nahrazení ze struktury nativního lektinu CV-IIL pro vyřešení fázového problému.

Abstract:

This bachelor thesis is devoted to protein crystallography and X-ray diffraction, which are used for discovering the structure of the studied enzyme - ornithine aminotransferase (OAT). The OAT is derived from pea and was expressed in Escherichia Coli, as a recombinant protein. The second studied protein is lectin CV-IIL from Chromobacterium violaceum,which was also expressed in Escherichia Coli. The work with the OAT is focused on the optimization of the purification procedure and the choice of suitable environment for crystallization trials and collection of high resolution diffraction data. For lectin CVII-L the 3D-structure of single-point mutant was solved (mutation of serin at position 22 by alanin) using molecular replacement with the structure of the native, lectin CVII-L to solve the phase problem.
