Modulation of cardiomyogenesis by opioid peptides

Abstract:

Endogenní opioidní peptidy, jako jsou dynorfiny a enkefaliny, hrají důležitou roli v regulaci mnoha biologických procesů, mezi které řadíme vývoj srdce a kardiovaskulární aktivitu. Signalizace endogenních opioidních peptidů je zprostředkována prostřednictvím jejich interakce s opioidními receptory, které jsou lokalizovány jak na cytoplazmatické membráně, tak na jaderné membráně. Některá data naznačují, že by dynorfiny a enkefaliny mohly napomáhat při regeneraci srdce, a to zejména prostřednictvím podpory regulace diferenciace srdečních kmenových buněk. Avšak účinky opioidních peptidů na kardiomyogenezi nebyly na buněčné a molekulární úrovni popsány natolik, aby mohly být využity v rámci terapie poškozeného srdečního svalu. Cílem diplomové práce bylo proto objasnit působení dynorfinů a enkefalinů na proces kardiomyogeneze in vitro. V rámci práce byl sledován efekt vybraných opioidních peptidů na diferenciaci myších embryonálních kmenových buněk do kardiomyocytů in vitro prostřednictvím tvorby embryoidních tělísek. Exprese genů specifických pro kardiomyocyty a exprese opioidních receptorů byly stanoveny pomocí metody kvantitativní RT-PCR, a na proteinové úrovni pomocí metody western blot, konfokální mikroskopie a průtokové cytometrie. Výsledky potvrdily, že použitý model je funkční, exprese genů specifických pro kardiomyogenní fenotyp (těžké řetězce myozinu β, Nkx2.5) je zvýšená a exprese genů charakteristických pro pluripotentní fenotyp buněk (Oct4, Nanog) je snížená. Nicméně, ovlivnění diferencujících se buněk opioidními peptidy v námi použitém modelu nemělo signifikantní vliv na průběh kardiomyogeneze. V rámci této práce byla vůbec poprvé popsána exprese opioidních receptorů, jejich buněčná lokalizace a změna jejich exprese v průběhu diferenciace myších embryonálních kmenových buněk do kardiomyocytů. Exprese opioidních receptorů se zvyšuje během kardiomyogeneze. Buněčná lokalizace byla pomocí imunocytochemie detekována především na jaderné membráně, méně na cytoplazmatické membráně. Tyto údaje o výskytu kappa a delta opioidních receptorů napomáhají k vysvětlení významu signalizace přes opioidní receptory v průběhu kardiomyogeneze in vitro a k všeobecnému ponětí o výskytu těchto receptorů. Jejich buněčná distribuce je také důležitá z hlediska zacílení léčiv v případném návrhu terapeutické aplikace a také k ověření vybraných metod a protilátek pro samotnou detekci opioidních receptorů u jiných buněčných linií a tkání nejen ve vztahu ke kardiomyogenezi, ale i v souvislosti s CNS. V závěru lze shrnout, že exprese opioidních receptorů se zvyšuje během diferenciace myších embryonálních kmenových buněk. Získané výsledky nepotvrdily významný vliv opioidních peptidů na kardiomyogenezi in vitro v námi použitém modelu, nejspíše z důvodu nízkého výskytu opioidních receptorů na buněčné membráně.

Abstract:

Endogenous opioid peptides such as dynorphins and enkephalins play an important role in regulation of many biological processes including heart development and cardiovascular function. Signaling of endogenous opioid peptides is mediated through their interaction with opioid receptors that are localized on the cytoplasmic membrane as well as the nuclear membrane. Interestingly, some data suggest that dynorphins and enkephalins could potentiate heart regeneration through stimulation of the cardiac stem cells regenerative potential. However, the effects of opioid peptides on cardiomyogenesis at cellular and molecular level are not well understood to be used for the therapy of heart muscle damage. Therefore, the aim of this diploma thesis was to clarify the effects of dynorphins and enkephalins during cardiomyogenesis in vitro. In this study, the action of selected opioid peptides was tested using the model of differentiation of mouse embryonic stem cells into cardiomyocytes in vitro via the formation of embryoid bodies. The expression of markers characterizing cardiomyogenesis and the expression of the opioid receptors were determined by quantitative RT-PCR and on the protein level by Western blot, by confocal microscopy and by flow cytometry. Results confirmed functionality of the employed model showing increased expressions of gene specific for cardiac phenotype (myosin heavy chains β, Nkx2.5) and decreased expressions of gene characteristic for the pluripotent phenotype of the cells (Oct4, Nanog). Nevertheless, supplementation of the differentiating cells by the opioid peptides did not show any significant effects on the course of cardiomyogenesis in our model. In this thesis, for the first time the expression of opioid receptors, their cellular localization and the change of their expression were described while differentiation of mouse embryonic stem cells into cardiomyocytes. The expression of opioid receptors increases during cardiomyogenesis. The cellular localization was detected by immunocytochemistry particularly on the nuclear membrane, less the cytoplasmic membrane. These data about the occurrence of kappa and delta opioid receptors contribute to the explanation of the signaling through opioid receptors during cardiomyogenesis in vitro and also the general idea about the occurrence of these receptors. Their cellular distribution is also important for targeting therapeutics in a possible design of therapeutic applications, and also to verify the selected methods and antibodies for the detection of opioid receptors for other cell lines and tissues, not only in relation to cardiomyogenesis, but also in relation to the CNS. In conclusion, the expression of opioid receptors increases during differentiation of mouse embryonic stem cells. These results did not confirm significant effects of opioid peptides on cardiomyogenesis in vitro in our model, probably due to the low occurrence of opioid receptors on the cell surface membrane.
