Structural characterization of an effect of heat stress on a organization of Photosystem II in the thylakoid membrane of higher plants using a circular dichroism

Abstract:

Tato diplomová práce je zaměřena na studium vlivu teplotního stresu na strukturní organizaci fotosystému II (PSII) v tylakoidních membránách vybraných druhů vyšších rostlin: huseníčku rolního, ječmene jarního a smrku ztepilého. Cílem této práce bylo zjistit strukturní a funkční změny PSII v tylakoidních membránách smrku indukované zvýšenou teplotou, jelikož dřívější studie prokázaly vyšší teplotní stabilitu fotochemie PSII u smrku oproti jiným druhům vyšších rostlin pěstovaných za stejných podmínek. Srovnáním výsledků měření teplotních závislostí fotochemické účinnosti PSII s výsledky měření teplotních závislostí cirkulárního dichroismu (CD) bylo ukázáno, že teplotní stabilita makroorganizace PSII všech studovaných rostlin souvisí s teplotní stabilitou fotochemie PSII. Pomocí teplotních závislostí CD spekter byla prokázána vyšší tepelná stabilita makrodomén u tylakoidních membrán smrku asi o 5 °C ve srovnání s membránami huseníčku a ječmene. Navíc pomocí velikostí amplitud hlavních psi-typu CD signálů bylo ukázáno, že smrk má nejspíš jinou organizaci LHCII v rámci PSII superkomplexů. CD spektra měřená na rozvolněných tylakoidních membránách vykazovaly stejný průběh u všech studovaných rostlin, takže excitonické interakce pigmentů ve světlosběrných komplexech se u těchto rostlin výrazně neliší. Při porovnání teplotních závislostí excitonických CD pásů nebo párů pásů bylo ale zjištěno, že i když k rozpadu trimerů na monomery dochází u všech rostlin při stejných teplotách, tak k rozpadu monomerů dochází u smrku při teplotách asi o 5 °C vyšších než u huseníčku a ječmene. Porovnáním teplotních závislostí CD pásů typu psi s výsledky nativní gelové elektroforézy uvedené ve studii Karlického a kol. (2016) bylo zjištěno, že zvýšená tepelná stabilita makrodoménové organizace pigment-proteinových komplexů v tylakoidní membráně u smrku je pravděpodobně spojena s udržením superkomplexů PSII v tylakoidní membráně při zvýšených teplotách.

Abstract:

This master thesis is aimed at studying the effect of heat stress on the structural organization of photosystem II (PSII) in thylakoid membranes of selected higher plant species: arabidopsis thaliana, spring barley and norway spruce. The aim of this study was to determine the structure and function changes of PSII in spruce thylakoid membranes induced by enhanced temperature, since earlier studies have shown higher thermal stability of PSII photochemistry for spruce compared to other species of higher plants grown under the same conditions. By comparing the results from the measurements of temperature dependence of PSII photochemical efficiency and of circular dichroism (CD), it was shown that thermal stability of PSII macro-organization of all studied plants correlates with the thermal stability of PSII photochemistry. Higher thermal stability in the macrodomains organization in spruce thylakoid membranes about 5 °C in comparison with Arabidopsis and barley ones was demonstrated by the temperature dependences of the CD spectra. Additionally, it has been reveled by the size of the amplitudes of the main psi-type CD signals that the spruce has probably different organization of LHCII within PSII supercomplexes. The CD spectra measured on unstacked thylakoid membranes showed the same pattern in all studied plants, and thus excitonic interactions of pigments within light-harvesting complexes do not differ significantly for these plants. However, comparison of the temperature dependences of excitonic CD bands or band pairs revealed that although the disintegration trimer into monomers occurs at the same temperatures in all plants, the disintegration of the monomers occurs in norway at temperatures of about 5 °C higher compared to Arabidopsis and barley. Further, comparison of the temperature dependences of CD psi-type bands with the results of native-gel electrophoresis indicated that the increased thermal stability of the macrodomains organization in spruce thylakoids is probably associated with maintaining of supercomplexes PSII in the tylakoid membrane at enhanced temperatures (Karlicky et al. 2016).
