The role of mitochondria in the physiology of cancer stem cells

Abstract:

Obecně je přijímáno, že součástí zhoubných nádorů je i populace málo diferencovaných nádorových buněk označovaných termínem nádorové kmenové buňky (cancer stem cells – CSCs). CSCs se vyznačují schopností iniciovat nádor a udržet jeho kontinuální růst, na základě čehož se CSCs přisuzují vlastnosti charakteristické pro kmenové buňky, tj. sebeobnova a potenciál diferencovat do vícero buněčných typů (ovšem v případě CSCs pouze do nádorových buněčných typů). CSCs jsou spojovány s progresí onkologického onemocnění, rozvojem metastáz a rekurencí nádoru. Poslední výsledky studií ukazují, že mitochondriální aktivita značně ovlivňuje vlastnosti a diferenciaci kmenových buněk. Vzhledem k podobnostem mezi kmenovými buňkami a CSCs se výzkum zaměřil i na úlohu mitochondrií ve fyziologii CSCs. Tato práce shrnuje aktuální poznatky o vztahu mitochondrií k fenotypu CSCs. Diskutována je rovněž funkce mitochondrií v odolnosti CSCs ke stresovým faktorům, respektive ke konvenční onkologické léčbě. Práce se věnuje také vlivu mitochondrií na proces metastazování. Závěrem jsou shrnuty látky, u kterých bylo zaznamenáno, že pozměňují vlastnosti mitochondrií nebo jejich aktivitu a mají terapeutický potenciál eliminovat CSCs.

Abstract:

It is widely accepted that malignant tumours also comprise a population of less differentiated cancer cells called cancer stem cells (CSCs). CSCs are characterized by the tumour initiating capacity and the ability to maintain continuous tumour growth. Thus, CSCs are assumed to possess stem cell-specific features, i.e. self-renewal and potential to differentiate into several cell types (in the context of CSCs only into cancer cell types). CSCs are associated with tumour progression, metastasis development and tumour recurrence. According to the latest studies mitochondrial activity potently affects the stem cell properties and differentiation. Regarding the similarities among stem cells and CSCs, scientists have focused on the role of mitochondria in the physiology of CSCs. This thesis summarizes recent findings concerning the link between mitochondria and CSC phenotype. Furthermore, it discusses the role of mitochondria in a resistance of CSCs to the stress conditions and cancer therapy. The thesis also deals with the contribution of mitochondria to the metastatic process. Finally, the thesis provides an overview of agents, which were reported to eliminate CSCs by the modulation of mitochondrial activity.
