Sequence-nonspecific interactions of protein p53 with DNA.

Abstract:

V literární části práce pojednává o proteinu p53, jeho struktuře a vazbě. Interakce s proteiny a různými formami DNA je mnohdy ovlivněna právě jeho strukturou. V důsledku jeho modifikací může docházet k inhibici jeho funkce a znemožnění vazby, nebo naopak k aktivaci latentního stavu proteinu a obnovení vazebné afinity. Povrchově je popsána vazba specifická, jelikož s vazbou nespecifickou může spolupracovat. Vzhledem k tomu, že nespecifická vazba je umožněna C-terminální doménou, v této práci je na ni kladen větší důraz. V experimentální části byla testována interakce specifická a nespecifická v závislosti na různé teplotě.

Abstract:

The literary part of the work deals with the p53 protein, it´s structure and binding. Interactions with proteins and various forms of DNA are very often influenced by it´s structure. As a result of the modifications can lead to inhibition of it´s function and disabling links or conversely to activate a latent state and restore protein binding afinity. In this work is specific binding described because of it´s cooperation with non-specific binding. Whereas, non-specific binding is made possible by the C-terminal domain is the greater emphasis. In the experimental part were tested interactions of specific and non-specific bindings depending on different temperature.
