Properties and clinical implications of de novo point mutations in human genetic disorders

Abstract:

Recentní sekvenační studie prokázaly, že se každý jedinec rodí na svět s asi 70 zárodečných de novo bodovými mutacemi. Podle mnoha studií patří zárodečné de novo bodové mutace mezi časté rizikové faktory vzniku mnoha genetických poruch a jejich vyšší četnost je spojována mimo jiné s pokročilým věkem otců při početí potomka. Tento nárůst se pohybuje okolo 1,5 variant s každým dalším rokem otce. Vznik nových sekvenčních variant s patologickými účinky může podmiňovat rozvoj vzácných onemocnění typu Kabukiho syndrom nebo Bohring-Opitz syndrom. Díky celoexomovému sekvenování je možné identifikovat asi 75 % těchto onemocnění. Častěji jsou však zárodečné de novo mutace zkoumány v souvislosti s běžnými neurovývojovými onemocněními. Typicky bývají spojovány s poruchami autistického spektra, epileptickými encefalopatiemi nebo intelektuálními poruchami. U těchto onemocnění je často pozorován nadbytek zárodečných de novo mutací v rizikových genech, které jsou exprimovány v různých částech mozkové tkáně. U výše zmíněných onemocnění a také u schizofrenie bylo objeveno 53 sdílených genů, které mohou pravděpodobně za podobné klinické příznaky poruch. Ačkoliv mají tyto poruchy podobnou etiologii, expresní vzory sdílených genů jsou pravděpodobně u jednotlivých onemocnění odlišné. Cílem této práce je popsat základní vlastnosti de novo bodových mutací, jejich původ, četnost, distribuci v rámci genomu a v neposlední řadě souvislost zárodečných de novo bodových mutací s genetickými onemocněními člověka.

Abstract:

Recent sequencing studies have shown that each individual is born with about 70 germline de novo point mutations. According to many studies, germline mutations are among the common risk factors for many genetic disorders. Their higher frequency is associated, among other factors, with the advanced age of fathers during the conception of the offspring. This increase is around 1.5 variants per every year of the father’s age. Creation of new pathological variants may induce development of rare diseases such as Kabuki syndrome or Bohring-Opitz syndrome. Thanks to whole exome sequencing, it is possible to identify about 75% of these diseases. Germline de novo mutations are more often investigated in connection with common neurodevelopmental diseases. They are typically associated with autism spectrum disorders, epileptic encephalopathies or intellectual disabilities. Patients with these diseases often suffer from an excessive incidence of germline de novo mutations in risk genes that are expressed in different parts of the brain tissue. There were 53 shared genes discovered between these diseases and schizophrenia which may probably cause similar symptoms. Although the etiology of these diseases is similar, expression patterns of the shared genes are probably different in each disease. The aim of this work is to describe features of de novo point mutations, their origin, rate, distribution over genome and last but not least the connection between germline de novo point mutations and human genetic diseases.
