Abstract:

ABSTRAKT DISERTAČNÍ PRÁCE Wild type (wt) protein p53 je významným nádorovým supresorem. Pro jeho správnou funkci je nepostradatelná spolupráce s dalšími proteiny p53 rodiny, p63 a p73. Úloha proteinů p63 a p73 v buňce nebyla dosud přesně zmapována. Mutace nebo nepřítomnost těchto proteinů v zárodečných buňkách vede k různým vývojovým vadám a deformacím, ale mohou působit i jako nádorové supresory. V některých případech p63/p73 částečně zastupují nefunkční p53 a spouštějí zástavu buněčného cyklu či apoptózu. Naopak jejich poškození negativně ovlivňuje funkci wt p53. Přesné zmapování vzájemných funkčních vazeb proteinů rodiny p53, signálních drah a objasnění jejich úloh v nádorové supresi může představovat další významný pokrok v onkologické léčbě. Látky podporující aktivitu wt p53 představují jeden z možných přístupů k léčbě nádorových onemocnění. Mezi ně patří i inhibitory cyklin dependentních kináz (CDK). Tyto látky zvyšují hladinu p53 v buňce, stimulují jeho aktivitu, a jsou tak schopny vyvolávat apoptózu u nádorových buněk. U některých inhibitorů CDK byla identifikována schopnost snižovat virovou replikaci a transkripci z virových promotorů. V případě, že by byly antivirové i protinádorové vlastnosti CDK inhibitorů potvrzeny dalšími nezávislými studiemi, představovala by tato skupina látek potenciální léčiva pro onkologické pacienty, která by s léčbou nádorového onemocnění zároveň zabraňovala šíření nebezpečných infekcí u oslabeného organismu. V předkládané práci jsem se věnovala třem tématům: 1. testování schopností izoforem p73 vázat se na promotory cílových genů wt p53, spouštět jejich transkripci a vyvolávat apoptózu nebo zástavu buněčného cyklu buněk nádorových buněčných linií. 2. studiu vztahů mezi izoformou DNp63a, ATM kinázou a fosforylací serinu 15 p53 u keratinocytové linie HaCat. 3. prokázání schopností nového inhibitoru CDK, olomoucinu II, potlačovat replikaci DNA-virů a silně inhibovat transkripci z různých virových promotorů.

Abstract:

DISSERTATION ABSTRACT The tumor suppressor p53 is critically important in the cellular damage response. The TP53-related genes, TP63 and TP73, exhibit significant structural homology to TP53, however, they do not code for classical tumor suppressors and are rarely mutated in human cancer. Both p63 and p73 exhibit tissue-specific roles in normal development and a complex contribution to tumorigenesis that is due to their expression as multiple protein isoforms. p63 and p73 isoforms regulate diverse cellular functions including proliferation, survival and differentiation. The net effect of p63/p73 depends on physical interactions between p63 and p73 isoforms, and on functional interaction with p53 at the promoters of specific downstream target genes. The multifaceted interactions occur in diverse ways in tumor-specific contexts, demonstrating a functional “p53 family signaling network” in human tumorigenesis. Understanding the role of p63 and p73 in the malignant transformation may ultimately provide new therapeutic opportunities for human cancer. Cell-cycle dysregulation is one of the cardinal characteristics of neoplastic cells. For this reason, small molecule inhibitors targeting cyclin-dependent kinases (CDKs) have been the focus of extensive interest in cancer therapy. In addition to inhibiting cell-cycle progression, these agents exhibit a variety of other activities, including interference with transcription and induction of apoptosis. CDKs are also required for the replication of viruses in dividing cells, such as adeno- and papillomaviruses and also for viruses replicating in non-dividing cells (HIV-1 and herpes simplex virus types 1 and 2). Pharmacological CDK inhibitors were shown to have potent antiviral activity in vitro and to inhibit specific functions of some viruses. Since tested CDK inhibitors prove to be non-toxic in human clinical trials, they may be useful in cancer treatment combining antiproliferative and antiviral properties. The presented study describes three different subjects: 1. the ability of p73 isoforms to bind on p53 responsive elements, to transactivate target genes and to regulate apoptosis and cell cycle arrest in tumor cell lines 2. the identification of a novel DNp63a -ATM-p53 signaling pathway active in human keratinocytes models as a UV damage –response mechanism in epithelial stem cells 3. the confirmation of inhibitory effect of olomoucine II and roscovitine on viral replication and transcription from viral promoters.
