Interactions of p53 proteins with DNA quadruplex

Abstract:

V této bakalářské práci se věnujeme studiu interakcí nádorových supresorových proteinů rodiny p53 se sekvencemi DNA schopnými tvorby guaninových tetraplexů (G­kvadruplexů). Tyto struktury se tvoří v biologicky významných oblastech genomu, mezi něž patří promotorové oblasti onkogenů. Proteiny rodiny p53 jsou nezbytné pro zachovávání integrity genomu regulací exprese cílových genů bránících vzniku nádorového bujení. Byly studovány interakce oligonukleotidů odpovídajících sekvencím G­bohatých promotorových oblastí genů c-Myc a hTERT se standardním proteinem p53 a centrálními doménami proteinů p53, p63 a p73 pomocí gelové retardační analýzy. Protein p53 i centrální domény p53, p63 a p73 se vázaly sekvenčně specificky na konsenzní sekvence v DNA. Předpokládáme, že pro vysoce afinitní strukturně selektivní vazbu proteinu p53 na DNA tvořící guaninový kvadruplex je důležitá především jeho C-koncová doména, jelikož protein p53CD tuto strukturu rozeznával s nižší afinitou. Interakce celého proteinu s kvadruplexovou DNA byla srovnatelná s vazbou sekvenčně specifickou.

Abstract:

In this thesis we studied interactions of p53 family of tumor supressor proteins with DNA sequences which can fold into guanine quadruplexes. G-quadruplexes can be formed in biologically relevant regions of the genome e.g. promoter regions of oncogenes. Proteins of the p53 family are essential for maintaining genome integrity, they inhibit tumor growth via regulation of expression of target genes involved in tumor supression. DNA-protein interactions of standard p53 protein and core domains of p53, p63 and p73 proteins with oligonucleotides which correspond to sequences of G-rich promoter regions of c-Myc and hTERT genes were studied by electrophoretic mobility shift assay. p53 protein and core domains of p53, p63 and p73 bound sequence-specifically to DNA consensus sequence. We suppose that high affinity structure­selective DNA binding of p53 to G-quadruplex is dependent on the C-terminal domain but also p53CD was able to bind G-quadruplex. Interaction of full lenght p53 protein with G­quadruplex DNA is comparably strong as sequence-specific binding.
