Coronaviruses and the host cell cycle

Abstract:

Koronaviry jsou původci některých respiračních onemocnění, která již několikrát vyústila v epidemii v různých oblastech světa. Aktuálním problémem je celosvětová pandemie onemocnění COVID-19, jehož původcem je virus ze skupiny beta-koronavirů SARS-CoV-2. Některé druhy mohou u člověka vyvolat těžké respirační problémy, které mohou vést až k úmrtí jedince. Za posledních 20 let se tedy zejména lidské koronaviry staly centrem výzkumu kvůli jejich vysoké patogenitě. Pochopení interakcí mezi koronaviry a hostitelskou buňkou je klíčové pro budoucí terapii či prevenci onemocnění způsobené touto skupinou virů. Interakce proteinů viru s buněčným cyklem hostitele ve většině případů vede k zastavení buněčného cyklu či k apoptóze, čímž si virus utváří adekvátní podmínky pro svou replikaci v hostitelské buňce. Od první epidemie v roce 2003 si koronaviry vyvinuly nové mechanismy pro manipulaci s buněčným cyklem a zvýšila se tak jejich komplexnost. Poznání těchto mechanismů nám může v budoucnu pomoct v predikci chování nových druhů viru a ve vývoji terapeutik.

Abstract:

Coronaviruses are the cause of some respiratory diseases that have resulted in epidemics several times in various parts of the world. A current problem is the worldwide pandemic of COVID-19, caused by the beta-coronavirus SARS-CoV-2. There could be some species that can cause severe respiratory problems in humans that can lead to the death of an individual. Thus in the last 20 years, human Coronaviruses have become the center of research due to their high pathogenicity in particular. Understanding the virus-host interaction is the key to the future therapy or prevention of diseases caused by this group of viruses. The interaction of virus proteins with the host cell cycle in most cases leads to cell cycle arrest or apoptosis, thereby creating adequate conditions for the virus replication in the host cell. Since the first epidemic in 2003, Coronaviruses have developed new mechanisms how to manipulate the host cell cycle, increasing their complexity. Knowledge of these mechanisms may help us in the future to predict the behavior of the new types of virus and also in the development of therapeutics.
