Problems of assessment mutagenicity in samples contaminated by polycyclic aromatic hydrocarbons

Abstract:

Cílem diplomové práce bylo provést hodnocení mutagenity vzorků čistírenských kalů Amesovým testem. Testovány byly dva vzory odvodněných anaerobně stabilizovaných nehygienizovaných kalů z ÚČOV Ostrava. K detekci mutagenní aktivity byly použity indikátorové kmeny Salmonella typhimurium His- TA98 a TA100. Test byl proveden ve variantách bez a s metabolickou aktivací in vitro jaterní mikrosomální frakcí S9 se směsí kofaktorů. Mutagenní aktivita byla detekována u obou vzorků na kmeni TA98 ve variantě testu s metabolickou aktivací in vitro. Tento výsledek indikuje přítomnost nepřímých posunových mutagenů v testovaných vzorcích. Nepřímá posunová mutagenní aktivita odpovídá vysokému obsahu PAU ve vzorcích detekovaných chemickou analýzou. Ve variantě testu s metabolickou aktivací in vitro na kmeni TA100, umožňujícím detekci mutagenů vyvolávajících substituce, byly oba vzorky vyhodnoceny jako potenciálně mutagenní. Varianta testu bez metabolické aktivace nemohla být do výsledků práce zahrnuta z důvodu vysokých hodnot spontánních revertant.

Abstract:

The aim of this study was to detect mutagenicity of sewage sludge using the Ames assay. Two samples of dewatered anaerobically stabilized and non-hygienized sewage sludges from the central sewerage plant in Ostrava were tested. Indicator strains Salmonella typhimurium His- TA98 and TA100 were used to the detection of frame-shift and substitute mutagenic activity with and without in vitro metabolic activation with S9 liver microsomal fraction including a cofactor mixture. Mutagenic activity was detected in both samples at the strain Salmonella typhimurium His- TA98 with metabolic activation in vitro. This result indicate the presence of indirect frame-shift mutagens in tested samples. Indirect frameshift mutagenic activity is in accord with high amount of PAHs detected by means of chemical analysis in the samples. On the base of results of the test with metabolic activation in vitro at the strain Salmonella typhimurium His- TA100 the samples are regarded as possible mutagenic. The variant of test without metabolic activation cannot be included in the results of this thesis because of a high count of spontaneous revertants.
