Modern molecular genetic technics in prenatal diagnosis of genetic deseases

Abstract:

Genetické změny na genové, chromozomové i genomové úrovni mohou být příčinou závažných onemocnění. Prenatální diagnostika umožňuje detekovat tyto choroby již ve fetálním stádiu, a poskytuje tak budoucím rodičům informace, na jejichž základě je možné rozhodovat o dalším průběhu těhotenství. Vyšetření genetického materiálu plodu je možné provádět různými přístupy. Klasickou metodou sloužící k prenatální diagnostice početních a strukturních změn chromozomů je karyotypování. Informace o primární struktuře DNA plodu, a tedy o abnormalitách na genové úrovni, lze získat pomocí polymerázové řetězové reakce a od ní odvozených technik. K prenatální diagnostice všech typů genetických onemocnění je možné využít i moderní molekulárně genetické přístupy, které mají potenciál přinášet podrobnější, přesnější a spolehlivější výsledky. Patří mezi ně například mikročipová varianta komparativní genomové hybridizace, kvantitativní fluorescenční polymerázová řetězová reakce a sekvenování nové generace. Cílem této práce je seznámit se s těmito technikami a popsat možnosti jejich využití v prenatální diagnostice.

Abstract:

Genetic changes at the single gene, chromosomal or whole genome level can cause very serious diseases. Prenatal diagnosis can detect these abnormalities in fetal period. That provides information about genetic state of the fetus, which enables the future parents to decide about further course of pregnancy. Examination of fetal genetic material can be performed by different approaches. The classical method used in prenatal diagnosis of numerical and structural chromosome changes is karyotyping. Information about primary structure of fetal DNA and thus about abnormalities at the single gene level can be obtained using polymerase chain reaction and its variants. For prenatal diagnosis of all types of genetic disorders, it is also possible to use modern molecular genetic technologies which have the potential to produce more detailed, accurate and reliable results. These include microarray-based comparative genomic hybridization, quantitative fluorescent polymerase chain reaction and next generation sequencing. The aim of this thesis is to describe these techniques and their possible use in prenatal diagnosis.
