Metabolomic analysis of human plasma by bio-inert UHPLC system coupled with mass spectrometry

Abstract:

Tato diplomová práce je zaměřená na vývoj a optimalizaci metody ultravysokoúčinné kapalinové chromatografie hydrofilních interakcí s hmotnostně spektrometrickou detekcí (HILIC-UHPLC/MS) a jejím využitím pro identifikaci metabolitů v lidské plazmě. Teoretická část je rozdělena na čtyři časti. První část se zabývá obecnými informacemi o metabolitech a jejich funkcí v lidském organismu. Druhá část je zaměřena na hmotnostní spektrometrií a bližší vysvětlení instrumentace používané v metabolomice. V třetí části jsou shrnuty všeobecné informace o kapalinové chromatografii se zaměřením na HILIC separaci. Poslední teoretická část je zaměřena na metabolomickou analýzu. V experimentální části jsou shrnuty výsledné postupy a použité parametry při analýze lidské plazmy a v závěrečné diskuzi jsou poté zhodnoceny a diskutovány výsledky této diplomové práce.

Abstract:

This thesis is focused on the development and optimization of the hydrophilic interaction ultrahigh-performance liquid chromatography with mass spectrometric detection (HILIC UHPLC/MS) and its use for the identification of metabolites in human plasma. The theoretical part is divided into four parts. The first part deals with general information about metabolites and their function in the human organism. The second part is focused on mass spectrometry and a closer explanation of the instrumentation used in metabolomics. The third part summarizes general information about liquid chromatography with a focus on HILIC separation. The last theoretical part is focused on the overall metabolomic analysis. In the experimental part, the resulting procedures and the parameters used in the analysis of human plasma are summarized, and in the final discussion, results of this diploma thesis are then evaluated and discussed.
