Evaluation and interpretation of the data obtained by the next-generation sequencing method

Abstract:

Tato bakalářská práce se zabývá metodami sekvenování nové generace. Charakterizuje vývoj technologií umožňujících stanovit pořadí jednotlivých bází a přibližuje metody masivního paralelního sekvenování s jejich výhodami i nevýhodami. Popisuje zásady důležité pro sekvenování nové generace a obsahuje postupy hodnocení výstupních dat. Součástí teoretické části je také popis genetických i negenetických příčin mentálních retardací. Praktická část je zaměřena na diagnostiku pacienta s projevy dětského autismu, středně těžkou mentální retardací a regresem ve vývoji pomocí platformy Illumina. Následuje ověření přítomnosti nalezené sekvenční varianty v exonu 15 genu HCFC1 u matky a bratra Sangerovým sekvenováním.

Abstract:

This bachelor thesis is dedicated to the next-generation sequencing methods. It characterizes the development of technologies able to determine the order of individual bases and approaches the methods of massive parallel sequencing with their advantages and disadvantages. It describes the principles that are important for next-generation sequencing and includes procedures used for evaluating output data. The genetic and non-genetic causes of mental retardation are also included in the theoretical part. The practical part is focused on the diagnosis of the patient with the symptoms of childhood autism, moderate mental retardation and regression in the development using the Illumina platform. Next step was verification of the sequence variant found in exon 15 of the HCFC1 gene of mother and brother with Sanger sequencing.
