Structural analysis of p53-binding sequence in supercoiled and linear DNA.

Abstract:

Významný nádorový supresor protein p53 je již od svého objevu intenzivně zkoumán díky své schopnosti regulovat buněčný cyklus a opravovat poškozenou DNA nebo vyvolat apoptózu. Díky těmto schopnostem protein zabraňuje vážnému poškození buněk a výskytu rakovinných onemocnění, které se vyskytují v případě absence či poškození proteinu. Aby byl protein schopný odhalit poškození buňky, je potřeba jeho navázání se na DNA. V této práci byly zkoumány sekvence DNA, u kterých by mohlo dojít k vazbě proteinu p53. K určení takových struktur byla v práci použita enzymatická sonda nukleáza S1 a restrikční endonukleázy.

Abstract:

A significant tumor suppressor the protein p53 has been since its discovery intensively studied for its ability to regulate cell cycle and repair DNA damage or induce apoptosis. With these capabilities this protein prevents serious damage to the cells and the occurrence of cancers that occur in the absence of or damage to the protein. The Protein was able to detect cell damage is a need for its binding to DNA. In this work, were examined for DNA sequence which could bind p53. To determine the structures were used in the work of an enzymatic probe S1 nuclease and restriction endonucleases.
