Metabolome of mountain gorillas (Gorilla beringei beringei) inhabiting Bwindi Impenetrable National Park, Uganda

Abstract:

Populace goril horských (Gorilla beringei beringei) v posledních desetiletích neustále roste a to zejména díky intenzivnímu ochranářskému úsilí ve Rwandě, Ugandě a Demokratické republice Kongo. Nicméně se objevují nové problémy, které tento druh nadále ohrožují. V populaci goril horských v Bwindi Impenetrable National Park (BINP) v Ugandě byly zaznamenány případy syndromu chronického chřadnutí (CWS). Nekropsie odhalily vysoký počet hlístic, konkrétně strongylidů, které způsobují střevní léze, jež mohou narušit střevní funkci a nakonec vést ke klinickému obrazu CWS. Vzhledem k tomu, že metabolické aktivity střevních mikrobů mohou výrazně ovlivnit fyziologii hostitele, v rámci mé práce jsem zkoumala metabolom goril v BINP. Cílem mé studie bylo získat přehled o tom, jak mohou změny ve společenstvech strongylidů modulovat gastrointestinální mikroekosystém a vysvětlit tak zhoršenou kondici těchto zvířat. Pomocí spektroskopie nukleární magnetické rezonance na frekvenci 500 MHz jsem ve vzorcích trusu goril identifikovala 20 metabolitů, včetně aminokyselin, karbonylových sloučenin, alkoholů, fenolů, esterů a organických kyselin. Množství kyseliny 3-(3-hydroxyfenyl)propionové, kyseliny 3-fenylpropionové a dvou mastných kyselin s rozvětveným řetězcem, konkrétně isobutyrátu a isovalerátu, se významně zvyšovalo s výskytem CWS pozitivních jedinců ve skupinách goril. Mé výsledky naznačují, že tyto metabolity mají proteolytický původ, což ukazuje na významnou korelaci mezi metabolickými změnami ve střevě a proteolytickou fermentací, jakož i na potenciální poškození střeva způsobené zejména strongylidy.

Abstract:

Mountain gorilla (Gorilla beringei beringei) populations have grown steadily in recent decades, thanks largely to intensive conservation efforts in Rwanda, Uganda and the Democratic Republic of Congo, but new challenges may be emerging further threatening this endangered species. Cases of chronic wasting syndrome (CWS) have been reported in the mountain gorilla population of Bwindi Impenetrable National Park (BINP) in Uganda. Necropsies revealed high numbers of nematodes, specifically strongylids, which cause intestinal lesions that can impair intestinal function and ultimately lead to the clinical presentation of CWS. As the metabolic activities of gut microbes can dramatically shape the physiology of the host, I explored the metabolome of Bwindi gorillas. The aim of my study was to gain insight into how changes in strongylid communities can modulate the gastrointestinal micro-ecosystem to explain their impaired fitness. Using 500 MHz nuclear magnetic resonance spectroscopy I identified 20 metabolites, including amino acids, carbonyl compounds, alcohols, phenols, esters, and organic acids in fecal samples of Bwindi gorillas. Abundances of 3-(3-hydroxyphenyl)propionic acid, 3-phenylpropionic acid, and two branched-chain fatty acids, namely isobutyrate, and isovalerate significantly increased with the prevalence of CWS-positive individuals in the gorilla groups. My results suggest that the metabolites have a proteolytic origin, indicating significant correlations between gut metabolic changes and proteolytic fermentation, as well as potential gut damage caused mainly by strongylids.
