The use of CRISPR/Cas9 for investigating targeted therapy opportunities in B-cell chronic lymphocytic leukemia
Abstract:
Chronická lymfocytární leukemie (CLL) je nejčastější leukemií dospělých a její výzkum je komplikován nedostatkem reprezentativních experimentálních modelů. Tato práce se zabývá tvorbou a charakterizací nových isogenních modelů CLL pomocí technologie CRISPR/Cas9. Byly připraveny buněčné linie nesoucí knockout nebo knock-in vybrané pro relevantní mutace (ATM, TP53, SF3B1, NOTCH1, MYD88, MS4A1) a ověřeny …more
Abstract:
Chronic lymphocytic leukemia (CLL) is the most common leukemia in adults, yet its research is limited by the scarcity of reliable experimental models. This thesis focuses on the development and characterization of novel isogenic CLL models using CRISPR/Cas9 technology. Cell lines carrying knockout or knock-in mutations in relevant genes (ATM, TP53, SF3B1, NOTCH1, MYD88, MS4A1) were established and …more
