Variability of selected cell-surface antigens in human embryonic stem cells

Abstract:

Human embryonic stem cells (hESC) acquire the properties of cancerously transformed cells during prolonged in vitro cultivation. The phenomenon of culture adaptation is a major hurdle for therapeutic application of hESC. The aim of this thesis is to determine the expression level of selected cell-surface antigens CD30, CD44, Cripto-1 (CR-1) typically found on cancerous cells. The different expression level of these antigens with diverse biological functions between low passage (unadapted) and high passage (adapted) cells implies various adaptive mechanisms. Changes in expression under different culture substratum emphasize the importance of microenviroment in modulating hESC behaviour as a response to the momentary state of microenviroment. This thesis also reveals an unpublished role of CR-1 in centrosomal abnormalities that represent the major mechanism leading to genomic instability associated with adapted hESC. Since CR-1 is involved in many signalling pathways, this finding suggests the complexity of mechanisms leading to centrosomal abnormalities.

Abstract:

Lidské embryonální kmenové buňky (hESC) získávají v průběhu kultivace in vitro charakteristiky nádorově transformovaných buněk. Tento proces adaptace představuje překážku pro využití hESC v medicíně. Cílem diplomové práce bylo zjistit úroveň exprese vybraných povrchových antigenů CD30, CD44, Cripto-1 (CR-1), které jsou ve zvýšené míře exprimované u nádorově transformovaných buněk. Rozdílná exprese těchto povrchových antigenů s odlišnými biologickými funkcemi mezi buňkami nízké (neadaptované hESC) a vysoké (adaptované hESC) pasáže poukazuje na různé adaptační mechanismy. Změny exprese v závislosti na kultivačním substrátu podtrhují význam mikroprostředí hESC v modulaci různých buněčných mechanizmů, které jsou odpovědí na okamžitý stav kultivačního prostředí. Výzkum odhalil doposud nepublikovaný vliv CR-1 na výskyt centrozomálních abnormalít, které jsou jedním z mechanismů vzniku genomové nestability typické právě pro adaptované hESC. Jelikož se jedná o molekulu, která je součástí mnohých signálních drah, centrozomální abnormality mohou vznikat v důsledku narušení rovnováhy rozličných buněčných pochodů.
