Characterization of retention with ternary mobile phases in HILIC chromatography

Abstract:

Diplomová práce je věnována výzkumu a optimalizaci podmínek ternárních (třísložkových) mobilních fází a gradientů v HILIC chromatografii (Hydrophilic Interaction Liquid Chromatography). Cílem bylo posoudit potenciální dopad kombinace methanolu s mobilními fázemi acetonitrilu a vodného pufru. Z recentní literatury vyplývá, že kombinace acetonitrilu a methanolu v mobilní fázi může ovlivňovat charakter difuzní vodné vrstvy na povrchu HILIC stacionární fáze a mít vliv na selektivitu separace. Jako modelové analyty byly zvoleny nukleové báze, nukleosidy, barbituráty a sulfonamidy, které patří do kategorie polárních látek vhodných pro separaci. Bylo proměřeno retenční chování v gradientech s acetonitrilem a methanolem a byly určeny parametry retenčního modelu z gradientových dat. Na základě parametrů predikovaných retenčních časů jsou diskutovány vlivy uvedených organických rozpouštědel.

Abstract:

The diploma thesis is devoted to the research and optimization of the conditions of ternary (three-component) mobile phases and gradients in HILIC chromatography (Hydrophilic Interaction Liquid Chromatography). The aim was to assess the potential impact of combining methanol with mobile phases of acetonitrile and aqueous buffer. Recent literature shows that the combination of acetonitrile and methanol in the mobile phase can influence the nature of the diffuse aqueous layer on the surface of the HILIC stationary phase and influence the selectivity of the separation. Nucleic bases, nucleosides, barbiturates and sulfonamides, which belong to the category of polar substances suitable for separation, were chosen as model analytes. The retention behavior in gradients with acetonitrile and methanol was measured and the parameters of the retention model were determined from the gradient data. Based on the parameters of the predicted retention times, the effects of the listed organic solvents are discussed.
