Investigation of cellular mechanisms to improve response to immunotherapy in B-lymphoid malignancies

Abstract:

Navzdory mnoha úspěchům v imunoterapii B-buněčných malignit, u velké části pacientů dochází ke ztrátě exprese cílových antigenů, což vede k rezistenci k léčbě. Identifikace mechanismů spojených se vznikem rezistence by tak mohla vést k navržení nových terapeutických možností a ke zvýšení efektivity léčby u těchto pacientů. Pro studium mechanismů zodpovědných za ztrátu cílového antigenu anti-CD20 monoklonálních protilátek byly vygenerovány rezistentní linie, u kterých byly důkladně analyzovány změny spojené se vznikem rezistence. Pro objasnění změn regulujících relaps po imunoterapii CAR-T lymfocyty byla vyizolována CD19 negativní linie z in vivo myšího modelu po relapsu, které byly dále analyzovány. Společně tyto poznatky povedou k lepšímu pochopení komplikací, které mohou vést k selhání léčby B-buněčných malignit.

Abstract:

Even though immunotherapy is successfully applied to treat B-cell malignancies, many patients relapse and become resistant to the treatment due to a target antigen loss. Identification of mechanisms underlying this phenomenon could lead to proposing novel therapy options and thereby to enhancing effectivity of treatment in these patients. To elucidate mechanisms accountable for the loss of anti-CD20 monoclonal antibody target antigen, we developed multiple resistant cell lines. The mechanisms connected with gained resistance of these cell lines were analysed. For the identification of changes underlying the relapse after CAR-T cell immunotherapy, CD19 negative cell line was isolated from an in vivo relapsed mouse model that was further analysed. Together, these findings lead to better understanding of complications that can cause therapy failure in B-cell malignancies.
