Expression analysis of gene LAG-3 in chronic lymphocytic leukemia cells

Abstract:

Chronická lymfocytární leukémie (CLL) je heterogenní a klinicky variabilní onemocnění a představuje nejčastější typ leukémie dospělého věku. Lymphocyte activation gene 3 (LAG3) je transmembránový protein nacházející se fyziologicky na T-lymfocytech, ale jeho exprese byla popsána na buňkách CLL s nemutovaným IGHV. Tato diplomová práce se zabývá expresí LAG3 v buňkách CLL v různých fázích onemocnění s důrazem na terapii. Průtokovou cytometrií byla analyzována exprese LAG3 na proteinové úrovni, pomocí kvantitativní PCR na úrovni mRNA a to u celkem 57 pacientů. Pomocí průtokové cytometrie byla exprese LAG3 ve všech případech detekována pouze na části populace CLL buněk. Na úrovni mRNA byla exprese LAG3 korelována s klinickým průběhem onemocnění a byl sledován vliv terapie na změny v expresi LAG3. Na základě výsledků našich analýz můžeme potvrdit, že v průběhu onemocnění, včetně období terapie, dochází ke změnám v expresi LAG3. Variabilita v expresi LAG3 odpovídá klinické heterogenitě CLL. Podrobnější analýza by mohla přispět k porozumění patogenezi CLL.

Abstract:

Chronic lymphocytic leukemia (CLL) is a heterogeneous and clinically variable disease and it is the most common type of adult leukemia. Lymphocyte activation gene 3 (LAG3) is a transmembrane protein found physiologically on T-lymphocytes, but its expression has been described on CLL cells with unmutated IGHV. This diploma thesis deals with the expression of LAG3 in CLL cells at various stages of the disease with an emphasis on therapy. Analysis of LAG3 expression at protein level was performed by using flow cytometry, at mRNA level by quantitative PCR. Total number of patients was 57. By flow cytometry, LAG3 expression was detected in all cases only in a part of the CLL cell population. At the mRNA level, LAG3 expression correlated with the clinical course of the disease and the effect of therapy on LAG3 expression changes was monitored. Based on the results of our analysis, we can confirm that there are changes in the LAG3 expression during the disease, including the treatment period. The variability in LAG3 expression corresponds with CLL clinical heterogeneity. More detailed analysis could contribute to the understanding of CLL pathology.
